Instructions for Use : Dissolve 52.3gms in 1000ml of distilled water. Gently heat to boiling with gentle swirling and dissolve the medium completely. DO NOT AUTOCLAVE. Cool to 45-50°C prior to dispense. Appearance: Greenish yellow colour, solution opalescent with flocculent precipitate pH (at 25°C): 7.7 ± 0.2 Principle : BISMUTH SULPHITE AGAR is used for the selective isolation of Salmonella spp. Salmonella constitute the most taxonomically complex group of bacteria among Enterobacteriaceae. Bismuth Sulphite Agar is a modification of Wilson and Blair formula. Also this medium favors use of larger inoculum as compared to other selective media, as it has unique inhibitory action towards gram-positive organisms and coliforms. Peptone and Beef extract serve as sources as carbon, nitrogen, vitamins and essential growth factors. Disodium phosphate acts as a buffereing agent. Dextrose is a fermentable carbohydrate source of the growth of microorganisms. Bismuth Sulphite Indicator and Brilliant Green are complementary, inhibiting to Gram-positive bacteria and coliforms, allowing Salmonella spp. to grow. Ferrous Sulphate is used for H2S production. When H2S is present, the iron incorporated in medium is precipitated, and positive cultures produce the characteristic brown to black color with metallic sheen. Agar is the solidifying agent.

Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. Cool to 45-50°C and aseptically add reconstituted contents of one vial of Oxytetra Selective Supplement. Mix well and pour into sterile Petri plates. Principles : Yeast extract provides essential growth nutrients. Dextrose acts as carbon and energy source. Low pH helps to reduce the bacterial flora. Oxytetracycline makes the medium more selective by inhibiting the growth of Lactobacilli encountered in milk and milk-products at low pH.

Sterilize by autoclaving at 15 psi (121°C) for 10 minutes. Cool to 45 - 50°C and pour into sterile Petri plates. Appearance: Red - brown colour, clear gel pH at 25°C: 6.5 ± 0.2 Principle BPL AGAR (BRILLIANT GREEN PHENOL RED LACTOSE AGAR) is used for isolation and identification of Salmonellae except Salmonella typhi in faeces, urine and milk and foodstuffs by separating lactose and sucrose negative bacteria. The Peptone provides the essential nutrient nitrogen for growth of microorganisms. Brilliant green in the medium inhibits accompanying microorganisms. Medium contains Lactose, whose degradation to acid is indicated by the pH indicator Phenol red, which changes its colour to yellow. The indicator exhibits a deep red colour in the alkaline range. Agar is a solidifying agent. Sodium chloride maintains the osmotic balance of the medium. The growth of the accompanying Gram-positive microbial flora, Salmonella typhi and Shigella is largely inhibited by brilliant green. ADAM (1966) recommended that 0.2 % sodium deoxychlolate should be added to the culture medium to inhibit the swarming of Proteus colonies.

Dispense into bottles or tubes and sterilize by autoclaving at 15 lbs pressure (121°C) for 15 minutes. For best results, the medium should be used on the day it is prepared, otherwise, it should be boiled or steamed for a few minutes and then cooled before use. Principle : Proteose peptone, HM infusion powder and BHI powder serve as sources of carbon, nitrogen, essential growth factors, amino acids and vitamins. Dextrose serves as a source of energy. Disodium phosphate helps in maintaining the buffering action of the medium whereas sodium chloride maintains the osmotic equilibrium of the medium. With the addition of 10 % defibrinated sheep blood, it is useful for isolation and cultivation of Histoplasma capsulatum
